Sequence-Specific Recognition of Double-Stranded DNA by Peptide Nucleic Acid Forming Double-Duplex Invasion Complex

چکیده
Peptide nucleic acid (PNA) is an analog of natural acids, where the sugar-phosphate backbone DNA replaced by electrostatically neutral N-(2-aminoethyl)glycine backbone. This unique peptide-based enables PNAs to form a very stable duplex with complementary acids via Watson–Crick base pairing since there no electrostatic repulsion between PNA and DNA·RNA. With this high affinity, have been used in wide range fields, from biological applications such as gene targeting, engineering probe sensor developments. In addition single-stranded DNA, can also recognize double-stranded (dsDNA) through formation double-duplex invasion complex. hard achieve other artificial expected be promising method dsDNA cellula or vivo does not require prior denaturation dsDNA. paper, we provide basic knowledge mainly focus on research invasion.

Double duplex invasion by peptide nucleic acid: a general principle for sequence-specific targeting of double-stranded DNA.
Pseudocomplementary PNAs containing diaminopurine.thiouracil base pairs have been prepared and are shown to bind with high specificity and efficiency to complementary targets in double-stranded DNA by a mechanism termed "double duplex invasion" in which the duplex is unwound and both DNA strands are targeted simultaneously, each by one of the two pseudocomplementary peptide nucleic acids (PNAs)...

The so-called acpcPNA system bears a peptide backbone consisting of 4'-substituted proline units with (2'R,4'R) configuration in an alternating combination with (2S)-amino-cyclopentane-(1S)-carboxylic acids. acpcPNA forms exceptionally stable hybrids with complementary DNA. We demonstrate herein (i) strand displacements by single-stranded DNA from acpcPNA-DNA hybrids, and by acpcPNA strands fro...
متن کاملTranscription-mediated binding of peptide nucleic acid (PNA) to double-stranded DNA: sequence-specific suicide transcription.
Peptide nucleic acid (PNA) forms sequence-specific (PNA)2/DNA triplexes with one strand of double-stranded DNA by strand invasion. When formed with the template strand of DNA such a (PNA)2/DNA triplex can arrest transcription elongation in vitro and can thus act as an anti-gene agent. One of the major obstacles to applying PNA as an anti-gene agent in vivo is that PNA strand invasion occurs at ...
متن کاملSequence-specific and Selective Recognition of Double-stranded RNAs over Single-stranded RNAs by Chemically Modified Peptide Nucleic Acids
RNAs are emerging as important biomarkers and therapeutic targets. Thus, there is great potential in developing chemical probes and therapeutic ligands for the recognition of RNA sequence and structure. Chemically modified Peptide Nucleic Acid (PNA) oligomers have been recently developed that can recognize RNA duplexes in a sequence-specific manner. PNAs are chemically stable with a neutral pep...
متن کاملSequence-specific recognition of double helical nucleic acids by proteins.
The base pairs in double helical nucleic acids have been compared to see how they can be recognized by proteins. We conclude that a single hydrogen bond is inadequate for uniquely identifying any particular base pair, as this leads to numerous degeneracies. However, using two hydrogen bonds, fidelity of base pair recognition may be achieved. We propose specific amino-acid side chain interaction...
